Medium for Axospirillum: nitrogen free broth.

g/L L-or DL Malic acid 5.0 K2HPO4 (di-potassium hydrogen orthophosphate) 0.5 MgSO4.7H2O 0.2 NaCl 0.1 CaCl2 0.02 FeEDTA(1.64% soln) 4.0mL Agar,(if preparing plates) 15.0

 

1) Trace element soln., see below 2.0mL

2) Bromothymol blue for broths(0.5%, w/v in 0.2M KOH soln.) 2.0mL or for plates Congo red soln.(0.25%, w/v in water) 15.0mL

3) Vitamin soln. OR use yeast extract( if nitrogen is required) 0.5g/L Deionised water make up to 1.0L Adujust pH to 6.8 with KOH, dissolve approx, 4g in 5mL of H2O.

Autoclave at 121℃ for 15 mins.

 

1) Trace Element solution Na2MoO4.2H2O 0.2 MnSO4.H2O 0.235 H3BO3 0.28 CuSO4.5H2O 0.008 ZnSO4.7H2O 0.024 Deionised water 1.0L

2) Bromothymol blue solution Dissolve 0.5g of bromothymol blue in 100mL of 0.2M KOH 3) Vitamin solution Biotin 0.01 Pyridoxin 0.02 Deionised water 20.0mL Dilute from above stock, 50 times and add 1mL to each litre of medium. Store in freezer.
